A trace amount of neutral glycerol is added to the stain to prevent methanol volatilization or oxidation, while also allowing for clearer staining of blood cells. It is commonly used for blood and cell smears, bone marrow smears, and bacterial staining. Cytoplasm appears red, nuclei and bacteria appear blue, and eosinophilic granules appear orange-red.
