La 4-diméthylaminopyridine, communément abrégée en DMAP, est un agent acylatant nucléophile exceptionnellement fort.Le groupe diméthylamine donneur d'électrons dans sa structure résonne avec l'anneau parent (anneau pyridine), activant fortement l'atome d'azote pour la substitution nucléophile. Il catalyse de manière significative l'acylation/l'estérification d'alcools et d'amines/acides à faible réactivité fortement stériles,avec une activité d'environ 104 à 106 fois celle de la pyridine.
