Pharmaceutical Intermediate 2-Chlorobenzaldehyde CAS 89-98-5 99.5% Purity Colorless Liquid

2-Chlorobenzaldehyde (CAS 89-98-5) is one of the workhorse aromatic aldehydes in pharmaceutical synthesis, the starting point for imidazoline antihypertensives, for 2-chlorobenzyl alcohol and for a range of Schiff base and hydrazone intermediates. In these routes the aldehyde group has to react cleanly and completely, so the parameters that matter are assay, the level of the corresponding carboxylic acid, and the absence of the 3- and 4-chloro isomers that would carry through to an isomeric impurity in the drug substance.

This pharmaceutical intermediate grade is produced by controlled chlorination and hydrolysis, then fractionally distilled under vacuum through a packed column. The distillation removes both the ring isomers and the oxidation products that form during storage, and it is why our specification can hold 99.5 percent assay with a maximum of 0.3 percent 2-chlorobenzoic acid and 0.2 percent total other chlorobenzaldehyde isomers, batch after batch.

• Assay 99.5% minimum by GC: predictable stoichiometry in condensation and reductive amination steps, less chance of an unreacted aldehyde impurity in the API
• Isomeric purity control: 3-chloro and 4-chloro isomers held below 0.2% in total, so no isomeric drug substance impurity is generated downstream
• 2-Chlorobenzoic acid 0.3% maximum: limits the acidic impurity that can carry through crystallisation and complicate the final purification
• Water 0.10% maximum: protects moisture sensitive Grignard and organometallic steps carried out in the same plant
• Colour 30 APHA maximum: clean, consistent liquid that does not darken on standing and does not require carbon treatment at the customer site
• Batch traceability from raw material: each drum links to a distillation record, GC chromatogram and a retained sample held for 24 months

Used as an intermediate in the synthesis of imidazoline type antihypertensives, in the preparation of 2-chlorobenzyl alcohol and 2-chlorobenzylamine, in hydrazone and Schiff base chemistry, and in the manufacture of pharmaceutical intermediates that require an ortho substituted benzaldehyde with defined isomeric purity.

The liquid should be charged under nitrogen where the reaction is moisture sensitive, and drums should be blanketed after each use because the aldehyde absorbs oxygen slowly and forms the corresponding acid. For process development work we supply 1 kg amber glass bottles so a route can be qualified before bulk quantities are committed.

Packed in 1 kg amber glass bottles for laboratory use, 25 kg plastic drums for pilot batches and 250 kg iron drums with epoxy phenolic lining for production. All containers are nitrogen blanketed and sealed with tamper evident closures, labelled with lot number, assay, water content and production date.

Store between 5 and 25 degrees C in a dry, well ventilated area away from oxidisers, bases and direct sunlight. The material is an irritant and should be handled with gloves and eye protection; a GHS format safety data sheet is issued with the first shipment and on revision.

Every batch is released with a Certificate of Analysis reporting assay, isomer distribution, 2-chlorobenzoic acid, water, acidity, colour, refractive index and density, together with the actual GC chromatogram. A 200 g retention sample is held for 24 months for traceability and dispute resolution.
