Pharmaceutical Intermediate 1-Ethynyl-1-Cyclohexanol 98% CAS 78-27-3 White Solid

1-Ethynyl-1-cyclohexanol is cyclohexanone with acetylene added across the carbonyl, which leaves a tertiary alcohol and a terminal alkyne on the same carbon. That combination is why formulators keep buying it: the alkyne reacts where you want it to, usually by addition or coupling, and the hydroxyl keeps the molecule soluble in the polar solvents a reaction needs.

We sell the standard 98% grade for synthesis work. It arrives as a white crystalline solid that melts in the high twenties, so on a warm day in the plant it is already liquid and can be charged by pump. The impurity we control hardest is residual cyclohexanone, because it carries through the following step and is not always easy to strip.

• 98% minimum by GC: reliable equivalents for routine acetylenic addition and coupling steps
• Residual cyclohexanone under 0.5%: no carry through into the next step where it would be hard to remove
• Melts around 30 degrees C: can be melted and pumped, or charged as solid, whichever suits the plant
• Water 0.3% maximum: no hydrolysis of moisture sensitive reagents in the same pot
• Low colour, white to off white: no dark product when the solid is redissolved

Coupling and addition chemistry on the alkyne, preparation of propargyl type derivatives, and as the alcohol component in esterification where the tertiary hydroxyl gives a sterically hindered ester that resists hydrolysis.

It can be charged as a melt at 40 to 45 degrees C through a heated line, or as solid flake broken from the drum. If you melt it, keep the hold time short and the temperature below 60 degrees C, since prolonged heating at higher temperature encourages colour and oligomer formation.

Store below 25 degrees C, dry and sealed, away from strong oxidisers. In hot climates keep it in a shaded store; above 30 degrees C the material is liquid and the drum must stay upright.
