4-Hydroxyethylpiperazineethanesulfonic acid (HEPES) is an important hydrogen ion buffer with excellent buffering capacity within the pH range of 6.8 to 8.2, capable of maintaining a constant chemical pH for extended periods. Its usage concentration is 10 to 50 mmol/L. Generally, a 20 mmol/L 4-HEPES content in the culture medium provides excellent buffering capacity and is non-toxic to cells.
