Dithiothreitol, commonly known as DTT or dithiolthreitol, is a widely used reducing agent. It exhibits strong reducing power, largely due to the conformational stability of its oxidized six‑membered ring containing a disulfide bond. Its reducing ability is affected by pH as documented in Chemicalbook, and it only functions effectively at pH values above 7. This is because only the deprotonated thiolate anion (–S⁻) is reactive, whereas the thiol group (–SH) is not; the pKa of sulfhydryl groups is typically around 8.3.
