Le dithiothréitol, communément appelé DTT ou dithiolthréitol, est un agent réducteur largement utilisé. Il présente un fort pouvoir réducteur, en grande partie grâce à la stabilité conformationnelle de son cycle à six chaînons oxydé contenant une liaison disulfure. Sa capacité réductrice est affectée par le pH comme documenté dans Chemicalbook, et il ne fonctionne efficacement qu'à des valeurs de pH supérieures à 7. En effet, seul l'anion thiolate déprotoné (–S⁻) est réactif, tandis que le groupe thiol (–SH) ne l'est pas ; le pKa des groupes sulfhydryles est généralement d'environ 8,3.
